摘要:
为阐明小鼠断奶前后胃组织中生长素(Ghrelin)和Ghrelin酰基转移酶(GOAT)基因表达和蛋白质分泌与胃功能发育之间的关系,选取新生BALB/c小鼠10窝,分别于14、21、28、35、42、49和56日龄时随机选取10只小鼠(每窝1只)处死,采集胃黏膜组织样和血液,定量分析基因表达、蛋白质水平和H+-K+-ATPase活性。结果表明,胃GOAT mRNA表达和蛋白质水平均受到断奶的影响,并首次发现断奶时胃GOAT基因表达和蛋白质水平显著下降,GOAT mRNA表达与胃H+-K+-ATPase活性成显著正相关。胃黏膜组织中生长抑素(SS)和胃泌素(Gastrin)基因mRNA表达在21、28和35日龄较低,随后逐渐增加,45日龄时显著升高(P<0.05)。断奶前后胃可能通过自身调节GOAT水平来适应饮食的变化,表明GOAT可能参与对胃功能发育,生长抑素可能对GOAT有抑制作用。
Abstract:
To investigate how the stomach modulates ghrelin production and secretion as well as ghrelin O-acyltransferase(GOAT)expression in pre- and post-weaning mice, the blood and gastric mucosal tissues of mice were collected for quantifying protein content and mRNA expression as well as H+-K+-ATPase. Newborn BALB/c mice were selected at 14 d, 21 d, 28 d, 35 d, 42 d, 49 d and 56 d of age respectively. The results revealed that weaning strongly regulated gastric GOAT mRNA expression and protein contents. For the first time, noticeable decreases were found in protein and mRNA levels of GOAT on weaning age, and GOAT mRNA expression was positively correlated with gastric H+-K+-ATPase activity. The mRNA expression levels of gastric somatostatin(SS)and gastrin rose from 21 d to 45 d of age, reaching a peak on 45 d of age(P<0.05). In conclusion, the stomach itself of pre- and post-weaning mice regulates ghrelin and GOAT production to adapt to diet change, and GOAT is involved in the gastric function development. SS down-regulates GOAT level.
