摘要:
以黄粉虫为原料,乙醇为提取剂,黄粉虫中黄酮类物质的提取率为指标, 采用超声波辅助-水浴加热的方法,在单因素(液料比、提取温度、提取时间、超声波功率和乙醇体积分数)试验的基础上设计响应面试验进行工艺优化,在不同条件(光照、温度、pH值、添加强氧化剂、添加还原剂和添加食品添加剂)下分析其稳定性。结果表明,黄粉虫中黄酮类物质最佳提取条件为:液料比50∶1(ml/g),提取温度60 ℃,提取时间41 min,超声波功率400 W,乙醇体积分数76%, 在该工艺条件下黄酮类物质的提取率为0.563 3%。稳定性分析结果表明,黄粉虫中的黄酮类物质受光照的影响较为明显,在过酸或过碱条件下会分解,在低含量(质量分数≤0.25%)强氧化剂作用下,黄酮类物质的稳定性良好,在一定范围内,还原剂亚硫酸钠与食品添加剂对其稳定性的影响不明显。研究结果可为黄粉虫中黄酮资源的开发提供理论依据。
Abstract:
Using Tenebrio molitor as raw material, ethanol as extraction agent, the extraction rate of flavonoids in Tenebrio molitor as index, the ultrasonic auxiliary-water bath heating method was adopted. Based on the single factor (liquid-to-material ratio, extraction temperature, extraction time, ultrasonic power and ethanol concentration) experiments, the response surface test was designed to optimize the process. Its stability was analyzed under different conditions (light, temperature, pH, strong oxidants, reducing agents and food additives). The optimal extraction conditions for flavonoids in mealworm were as follows: liquid-to-material ratio was 50∶1 (ml/g), extraction temperature was 60 ℃, extraction time was 41 min, ultrasonic power was 400 W, ethanol concentration was 76%. Under these conditions, the extraction rate of flavonoids was 0.563 3%. The results of stability analysis showed that the flavonoids in mealworm were more obviously affected by light, and would decompose under the condition of acid or alkali. Under the action of strong oxidant with low content (≤0.25%), the stability of flavonoids was good. In a certain range, the effect of reducing agent sodium sulfite and food additives on its stability was not obvious. These results can provide a theoretical basis for the development of flavonoid resources in Tenebrio molitor.
