以B5为褐飞虱抗性供体亲本,以粳稻品种(品系)嘉隆17B和嘉58为受体亲本,通过杂交、回交和标记选择,获得Bph14-嘉隆17B、Bph15-嘉隆17B、Bph14/Bph15 -嘉隆17B及Bph14 -嘉58、Bph15-嘉58、Bph14/Bph15-嘉58导入系。苗期和抽穗期抗性鉴定结果显示,在苗期Bph14/Bph15-嘉隆17B或Bph14/Bph15-嘉58的抗性级别为1.0级(高抗),Bph14 -嘉隆17B或Bph14 -嘉58、Bph15 -嘉隆17B或Bph15 -嘉58抗性为 3.0~7.0级(抗、中抗、中感),双基因导入系抗性显著高于单基因导入系; 同时发现Bph14-嘉隆17B抗性水平(4.56级)低于Bph14-嘉58(2.50级),Bph15-嘉隆17B抗性水平(3.80级)高于Bph15-嘉58(5.86级); 抽穗期的抗性水平与苗期表现一致或接近。说明Bph14 、Bph15在粳稻背景中能够表达,但表达水平因背景不同而有差异,水稻苗期的抗性可作为抗性品种选育的参考依据,利用分子标记技术辅助选育抗褐飞虱粳稻品种是切实可行的。
Abstract:
A rice line B5, with Bph14 and Bph15 genes controlling the resistance to brown planthopper(BPH), was crossed with Jialong17B and Jia58, two Japonica varieties developed in Zhejiang province. The introgression lines, Bph14-Jia58, Bph15-Jia58, Bph14/Bph15-Jia58 and Bph14-Xiushui134, Bph15-Xiushui134, Bph14/Bph15-Xiushui134, were obtained via crossing, backcrossing and marker-assisted selection. The resistances to BPH were higher in double-gene introgression lines Bph14/Bph15- Jialong17B and Bph14/Bph15- Jia58(highly resistant)than those in single gene introgression lines(resistant, moderately resistant and moderately susceptible). Line Bph14-Jia58 was more resistant than line Bph14-Jialong17B, but line Bph15-Jialong17B was more resistant than line Bph15-Jia58 at seedling stage. At heading stage, the resistance levels of most introgression lines were consistent with those at seedling stage. This study indicates that Bph14 and Bph15 genes from B5 could be expressed in Japonica, and expression levels varied with the background. The selection of BPH-resistance could be conducted at seedling stage for marker-assisted selection of BPH-resistant Japonica.
