摘要:
为比较玉竹多糖、桑叶多糖、杜仲多糖、商陆多糖、淫羊藿多糖体外增强免疫活性,采用酶学检测法测定各中药多糖对鸡胚成纤维细胞的安全浓度,根据安全浓度测定结果,选择250.000 μg/ml、125.000 μg/ml、62.500 μg/ml、31.250 μg/ml、15.625 μg/ml 5个浓度,用MTT法比较5种中药多糖单独和PHA协同刺激对外周血T淋巴细胞增殖指数的影响,以及比较5种中药多糖单独和LPS协同刺激对脾脏B淋巴细胞增殖率的影响。结果显示, 玉竹多糖、桑叶多糖、杜仲多糖、商陆多糖、淫羊藿多糖对鸡胚成纤维细胞的安全浓度分别为1 250.0 μg/ml、625.0 μg/ml、625.0 μg/ml、625.0 μg/ml、312.5 μg/ml。玉竹多糖、桑叶多糖、杜仲多糖和淫羊藿多糖无论是单独还是协同PHA刺激外周血T淋巴细胞,均不同程度地表现出增强T淋巴细胞的活性。桑叶多糖、杜仲多糖和淫羊藿多糖无论是单独还是协同LPS刺激脾脏B淋巴细胞,均不同程度地显示出良好的增强B淋巴细胞的活性。桑叶多糖和杜仲多糖体外增强免疫效果最佳,在62.500 μg/ml和125.000 μg/ml时,均显示出较强的增强外周血T淋巴细胞和脾脏B淋巴细胞增殖的能力。
Abstract:
Polygonatum odoratum polysaccharide (POPS), morus polysaccharide(MPS), eucommia polysaccharide(EPS), pokeweed polysaccharide (PPS), and epimedium polysaccharide(EPPS) to chicken embryonic fibroblast(CEF) in vitro were determined and showed as follows compared. 1 250.0 μg/ml, 625.0 μg/ml, 625.0 μg/ml, 625.0 μg/ml, and 312.5 μg/ml, respectively. The effects of above five polysaccharides applied solely or co-applied with phytohaema glutinin(PHA) on the proliferation of peripheral blood T lymphocytes and the effects of five paly saccharides appiied solely or co-applied with lipopolysaccharide LPS on the proliferation of spleenic B lymphocytes were compared. POPS, MPS, EPS and EPPS could enhance peripheral blood Tlymphocytes activity at different levels either stimulated alone or co-stimulated with PHA. MPS, EPS and EPPS could improve spleenic B lymphocytes activity at different levels either stimulated alone or co-stimulated with LPS. The immune enhancements of MPS and EPS in vitro outperformed others at the concentration of 62.5 μg/ml and 125 μg/ml.
