Abstract:
Due to the high genetic variability of porcine reproductive and respiratory syndrome virus (PRRSV) and its complex immune evasion mechanisms, existing vaccines have certain limitations in cross-protection, making antiviral drugs an important complementary strategy for preventing PRRSV infection in swine production. This paper takes the antiviral mechanism of PRRSV as its main logical thread and systematically reviews research progress on six major strategies: targeting viral entry, including receptor blocking and endocytosis inhibition; targeting viral replication, including RNA-dependent RNA polymerase (RdRp) and protease inhibitors; modulating host innate immunity, including interferon and Toll-like receptor/stimulator of interferon genes (TLR/STING) pathway agonists; intervening in host metabolism and stress, including lipid metabolism and oxidative stress regulation; gene silencing and antisense oligonucleotide technologies, and innovative biologics, including nanobodies and miniproteins. At the mechanistic level, this review provides an in-depth analysis of the multi-level intervention targeting the CD163 receptor, the inhibition mechanisms of viral proteases and polymerases, and the balance between innate immune activation and inflammatory regulation. In terms of future prospects, this paper proposes four major development pathways: multi-target synergistic intervention, systematic exploration of drug repurposing, nano-delivery-empowered natural products, and AI-driven drug design. The findings of this review can provide a theoretical reference for the further development and clinical application of antiviral drugs against PRRSV.
