摘要:
以欧前胡素为内标物,建立了测定川白芷药材中白当归素、佛手柑内酯、氧化前胡素、欧前胡素、异欧前胡素多种成分含量的一测多评方法。采用HPLC方法,色谱柱为ZORBAX Eclipse Plus C18 (4.6 mm×250.0 mm,5 μm),以甲醇-水为流动相,等度洗脱,柱温30 ℃,检测波长为250 nm,流速为1 ml/min进行测定。以欧前胡素为内标物,建立川白芷药材中白当归素、佛手柑内酯、氧化前胡素、异欧前胡素的相对校正因子。川白芷药材中白当归素、佛手柑内酯、氧化前胡素、异欧前胡素的相对校正因子重现性良好,分别为0.407、0.844、0.847、0.625,一测多评法测定结果与外标法测定结果无显著差异。表明以欧前胡素为内标物建立川白芷药材多种成分的一测多评法测定结果准确性良好,方法可行,可作为川白芷药材的质量控制方法。
Abstract:
The quantitative analysis of multi-components by single-marker (QAMS) method for content determination of byakangelicin, bergapten, oxypeucedanin, imperatorin and isoimperatorin in Angelica dahurica was established by setting imperatorin as internal standard substance. The high performance liquid chromatography (HPLC) method was adopted, and the chromatographic column of ZORBAX Eclipse Plus C18 ( 4.6 mm×250.0 mm, 5 μm) was used. The mobile phase was composed of methanol and water in isocratic elution. The column temperature was controlled at 30 ℃, the flow speed was 1 ml/min, and the detection wavelength was 250 nm. The relative correction factors of imperatorin to other four components were established by setting imperatorin as internal standard substance. The relative correction factors of byakangelicin, bergapten, oxypeucedanin and isoimperatorin in Angelica dahurica were 0.407, 0.844, 0.847 and 0.625, which had a high reproducibility. There was no significant difference between the results of QAMS method and external standard method. The QAMS method established by setting imperatorin as internal standard substance is accurate and feasible, which can be used as a useful quality control method for Angelica dahurica.
