摘要:
利用简单序列重复区间扩增多态性(ISSR)技术分析了罗汉果航天、无籽、野生、栽培4类种质的遗传差异,构建DNA指纹图谱和UPGMA聚类图,并进行杂交优势预测的应用研究。结果表明,筛选出的19个ISSR引物中的5个可直接鉴别罗汉果21份特异种质,种质间的遗传相似性系数为 0.656~0.896,在相似性系数为0.795处的聚类结果分为5类。不同种质间存在一定的遗传变异,尤其是广西金秀(S17)、湖南永州(S18)和广西百色(S19)的野生种质与桂林主产区种质间,航天种质内以及雌雄株间都产生了较大的遗传差异。根据遗传差异的大小,对9个雌株预测了20个可能获得理想杂交优势的组合,推测雄株S13、S7和S9是较好的父本材料。该研究补充完善了罗汉果种质资源的遗传背景信息,从而为罗汉果种质的保护、创新和利用提供了理论基础及依据。
Abstract:
The genetic diversity of four lines of Siraitia grosvenorii germplasms (aerospace, seedless, natural and cultivated) was analyzed by inter-simple sequence repeat (ISSR), the DNA fingerprints and UPGMA dendrogram were established, and the heterosis was predicted. The results showed that five out of 19 selected ISSR primers could directly identify all 21 S. grosvenorii specific germplasms. Genetic similarity coefficients ranged from 0.656 to 0.896, and the germplasms were divided into five groups at 0.795. It revealed genetic diversity among different germplasms. Particularly, larger diversity existed among the germplasms from different geographic areas, i.e. Hunan Yongzhou(S18), Guangxi Baise(S19), Guangxi Jinxiu(S17) and the major-producing area of Guilin, as well as within the aerospace germplasms, between male and female plants. Based on the genetic diversity, we deduced 20 parental combinations of nine female individuals, and aerospace S13, natural S7 and S9 were more excellent male parent. The research complemented new genetic background information of S. grosvenorii and would provide the theoretical basis and reference for conservation, innovation and utilization of these germplasms.
