随着动物源性产品(农产品、食品和饲料)中掺假造假事件不断增多以及动物来源性疾病的传播风险逐渐加大,作为保障食品安全和维护消费者权益的高效检测手段,动物源性成分的定性和定量检测技术已成为国内外的研究热点。本研究通过比对分析8种动物的线粒体全基因组序列筛选出羊源性成分特异性的引物和探针组合进行基于实时荧光PCR技术的羊源性成分鉴定。结果表明,所研发的羊源性成分检测的引物和探针具有高度的特异性,同时具有较好的灵敏度(可检测到1 pg的DNA)。羊肉制品中羊源性定量检测试验结果说明,此方法具有较好的定量检测能力。综上所述,以此引物和探针为基础的检测方法适用于羊源性食品和农畜产品的动物源性成分检测。

Due to the increasing of complaints for adulterated foods and feeds and propagation risk of diseases derived from several animals, the qualitative and quantitative detection techniques of animal-derived substances in foods and feeds are significant biotechnical methods to ensure food safety and protect consumer rights. To design the primer and probe with intellectual property which could be used to detect sheep-derived materials by means of TaqMan real-time PCR, the complete mitochondrial genome sequences of eight animals were compared. The results showed that the technique based on the primers and probe was specific for species identification and quantification, and had good sensitivity. The results of quantitative detection of sheep-derived material showed that the method had good quantitative detection ability. This biotechnological method based on TaqMan real-time PCR was appropriate to identify sheep-derived materials in farm products and foods.
