摘要:
为建立快速、简便检测猪瘟病毒(CSFV)抗体的方法,本研究采用胶体金和量子点免疫层析技术,将纯化的重组CSFV E2蛋白作为捕捉抗原,以纯化的抗CSFV E2蛋白多克隆抗体和HRP兔抗猪IgG分别包被于硝酸纤维素膜上,作为质控线(C线)和检测线(T线),优化反应条件,制备免疫层析试纸条,用于临床检测。结果表明,制备的2种试纸条都可用于检测CSFV标准阳性血清,使用不同批次试纸条进行检测,结果无差异。2种试纸条与CSFV阳性血清反应呈阳性,与伪狂犬病毒(PRV)、猪乙型脑炎病毒(JEV)、猪繁殖与呼吸综合征病毒(PRRSV)和圆环病毒2型(PCV2)反应呈阴性;胶体金试纸条在临床样品中阳性检出率为84.38%(81/96),与CSFV抗体检测ELISA试剂盒检测阳性符合率为92.05%(81/88);量子点试纸条阳性检出率为87.50%(84/96),与ELISA试剂盒检测阳性符合率为95.45%(84/88)。本研究制备的2种试纸条都具有较好的特异性、敏感性、重复性和稳定性。
Abstract:
Two test strips for rapid detection of the classical swine fever virus(CSFV) antibody were developed based on colloidal gold and quantum dots immunochromatographic assay. The recombinant CSFV E2 protein was incubated on test strip as capture antigen, and the rabbit anti-pig IgG was immobilized on the test line as detection antibody. In addition, anti-CSFV E2 protein polyclonal antibody was blotted on the control line of the nitrocellulose membrane. The results showed that the two test strips could be used to detect the CSFV positive serum. The test strips did not capture unspecific virus proteins which derived from pseudorabies virus(PRV), Japanese encephalitis virus(JEV), porcine reproductive and respiratory syndrome virus(PRRSV) and porcine circovirus type 2(PCV2). In the detection of clinical samples, the positive detection rate of colloidal gold immunochromatographic strip was 84.38% (81/96), and the coincidence rate was 92.05%(81/88) compared with CSFV antibody IDEXX kit (ELISA). The positive detection rate of quantum dot immunochromatographic strip was 87.50%(84/96), and the coincidence rate was 95.45%(84/88)compared with CSFV antibody IDEXX kit (ELISA). The two test strips prepared in this study have good specificity, sensitivity, repeatability and stability.
