摘要:
为评价高效佐剂VA5对猪细小病毒和猪圆环病毒二联灭活疫苗的免疫增强效果,将VA5与猪细小病毒、猪圆环病毒疫苗混合免疫BALB/c 小鼠和豚鼠。结果显示,VA5佐剂能明显提高猪细小病毒血凝抑制抗体效价和血清病毒中和抗体效价,亦能提高猪圆环病毒ELISA抗体和中和抗体效价,并可降低二种抗原的使用量。VA5佐剂显著提高小鼠和豚鼠免疫后的淋巴细胞刺激指数。攻毒后,VA5佐剂处理组动物脾脏PCV2/PPV病毒载量明显低于不含高效佐剂的免疫组。 VA5佐剂能提高猪细小病毒、猪圆环病毒疫苗在小鼠/豚鼠中的抗体效价,提高细胞免疫力,降低攻毒后的排毒。
Abstract:
To evaluate the effect of immunopotentiator VA5 on the porcine parvovirus (PPV) and porcine circovirus type 2 (PCV2) bivalent inactivated vaccine (PPV-PCV2), BALB/c mice and guinea pigs were immunized the bivalent vaccine with or without VA5. The results showed that VA5 significantly improved the serum-virus neutralizing (SN) and hemagglutination inhibition antibo-dies titer against PPV, and similarly increased the ELISA antibody levels and SN antibody levels against PCV2. The VA5 significantly reduced antigen contents in each dose of vaccine. Lymphocyte stimulation index of mice and guinea pigs were significantly enhanced in the treatment of bivalent vaccine plus VA5. After challenge with PCV2/PPV, the viral load of PCV2/PPV in the spleen of mice and guinea pigs were both significantly reduced in the group immunized with the bivalent vaccine in presence of VA5 compared with that in the group immunized with bivalent vaccine absence of VA5. VA5 adjuvant can improve antibody titer of the porcine parvovirus, porcine circovirus vaccine in mice/guinea pig, improve cellular immunity and reduce the detoxification after attack.
