摘要:
为探讨党参多糖(CPP)对仔猪小肠黏膜免疫功能的影响,选取60头1日龄健康“杜×长×大”三元杂交仔猪,随机分为3组,从14 日龄开始,3组仔猪分别饲喂代乳料(对照组)、代乳料+1%(以生药计)CPP(低剂量组)、代乳料+2% CPP(高剂量组)。21 日龄断奶,试验期为14 d。试验结束时,每组随机选择6头仔猪进行剖杀,取小肠各段分别放置于4%多聚甲醛和液氮中保存,免疫组化法(IHC)检测分泌型免疫球蛋白A(SIgA)、免疫球蛋白G(IgG)和免疫球蛋白M(IgM)阳性表达水平,逆转录聚合酶链式反应(RT-PCR)测定IgA、IgG和IgM的 mRNA相对表达量。结果表明党参多糖显著或极显著提高仔猪小肠SIgA、IgG和IgM蛋白表达水平以及IgA、IgG和IgM mRNA相对表达量(P<0.01或P<0.05),其中添加2%党参多糖的效果优于添加1%党参多糖的效果。表明在仔猪饲粮中添加党参多糖能够促进仔猪小肠IgA、IgG、IgM mRNA表达,提高仔猪机体免疫应答能力。
Abstract:
This experiment was conducted to analyze the effect of supplementary Codonopsis pilosula polysaccharide (CPP) on small intestinal mucosal immunity of piglets. Sixty crossbred piglets “Duroc × Landrace × Yorkshire” (1-day-old) were randomly divided into three groups. Piglets were fed with milk replacer, milk replacer plus 1%(dry herbs)CPP and milk replacer plus 2% CPP from 14-day-old in control group, low dosage group and high dosage group, respectively. All piglets were weaned at 21 d. The trial was conducted for 14 d. At the end of trial, six piglets from each group were slaughtered. Each segment of the small intestine was collected and placed in 4% paraformaldehyde and liquid nitrogen. The protein positive expression levels of secretory immunoglobulin A (SIgA), immunoglobulin G (IgG) and immunoglobulin M (IgM) were measured by immunohistochemistry (IHC), and the relative expression of IgA, IgG and IgM mRNA were measured by RT-PCR. The results showed that supplementary CPP could elevate significantly or very significantly (P<0.01 or P<0.05) the protein expression levels of SIgA, IgG and IgM and the relative expression of IgA, IgG and IgM mRNA in the small intestine. The adding effect of 2% supplementary CPP was greater than that of 1% supplementary CPP. Supplementary CPP could promote the IgA, IgM, IgG mRNA transcription of small intestine, and enhance immune response in piglets.
