摘要:
为探究复配了其他药食同源植物粉的葛根全粉制品的体内外抗氧化活性及抗衰老功效,本研究基于网络药理学筛选出了葛根、甘草、马齿苋、金银花、菊花等药食同源植物,用葛根全粉与其他药食同源植物粉复配加工成复配了其他药食同源植物粉的葛根全粉制品(LPH1、LPH2)。以自由基清除率为评价指标,测定其体外抗氧化活性;并探究不同剂量(12.5%、25.0%、50.0%)的复配了其他药食同源植物粉的葛根全粉制品对果蝇存活时间、攀爬能力、体内抗氧化酶活性及氧化应激耐受能力的影响。结果表明,LPH1处理与LPH2处理对DPPH自由基、ABTS·+的清除率均显著高于葛根全粉原料对照(CK)(P<0.05)。果蝇活体试验结果显示,25%为复配了其他药食同源植物粉的葛根全粉制品最优添加剂量,与对照相比,此剂量下LPH1处理、LPH2处理果蝇的平均寿命分别显著延长25.84%和26.41%;且在H2O2与百草枯氧化应激模型中,果蝇的存活时间均得到显著延长(P<0.05)。综上,复配了其他药食同源植物粉的葛根全粉制品可有效提升果蝇体内外抗氧化活性、延缓其衰老进程,本研究结果可为新型葛根全粉制品研发提供理论依据。
Abstract:
To investigate the in vitro and in vivo antioxidant activity and anti-aging effects of Pueraria lobata whole powder products blended with other medicine-food homologous plant powders, this study screened medicine-food homologous plants, including kudzu root (Pueraria lobata), licorice (Glycyrrhiza uralensis), purslane (Portulaca oleracea), honeysuckle (Lonicera japonica), and chrysanthemum (Chrysanthemum morifolium), based on network pharmacology. These plants were processed with kudzu root whole powder to formulate the kudzu root whole powder products blended with other medicine-food homologous plant powders (designated as LPH1 and LPH2). Using free radical scavenging rate as the evaluation index, the in vitro antioxidant activity of the products was determined. Furthermore, the effects of different doses (12.5%, 25.0%, and 50.0%) of the kudzu root whole powder products blended with other medicine-food homologous plant powders on the survival time, climbing ability, in vivo antioxidant enzyme activities, and oxidative stress tolerance of Drosophila melanogaster were investigated. The results showed that the scavenging rates of DPPH and ABTS·+ radicals in both LPH1 and LPH2 treatments were significantly higher than those in control (CK) (P<0.05). The results of the in vivo Drosophila melanogaster assay indicated that 25% was the optimal concentration of the kudzu root whole powder products blended with other medicine-food homologous plant powders. At this concentration, compared with the control, the mean lifespans of Drosophila melanogaster in LPH1 and LPH2 treatments were significantly extended by 25.84% and 26.41%, respectively. In the H2O2 and paraquat-induced oxidative stress models, the survival time of Drosophila melanogaster was significantly extended (P < 0.05). In conclusion, the kudzu root whole powder products blended with other medicine-food homologous plant powders effectively enhanced the in vitro and in vivo antioxidant activity of Drosophila melanogaster and delayed their aging process. These findings of this study provide a theoretical basis for the development of novel kudzu root whole powder products.
